Serum Inhibition of Plasmin

چکیده

In the various studies tha t have been done on the inhibition of proteolytic enzymes by serum, no distinction has been made in the inhibition of different enzymes. I t has generally been assumed that all proteolytic inhibition is caused by the same substance in serum and that this substance is occasionally increased in association with disease. There has been no evidence for the existence of separate proteolytic inhibitors. There have been several at tempts to isolate the material in serum responsible for the inhibition of proteolytic enzymes (1, 2), but no inhibitory material has yet been obtained in pure form. The chemical characteristics of the inhibitory material in serum are not well known. The data t0 be presented indicate tha t more than one antiproteolytic factor can be detected in serum and that these factors act selectively on the enzymes plasmin, trypsin, and chymotrypsin. For the sake of convenience, the factor in serum which inhibits an enzyme will be referred to as the enzyme inhibitor (e.g., plasm{n inhibitor, trypsin inhibitor), although each inhibitor m a y b e composed of several substances or groups of substances.

THE INHIBITION OF PLASMIN, PLASMA KALLIKREIN, PLASMA PERMEABILITY FACTOR, AND THE C'1r SUBCOMPONENT OF THE FIRST COMPONENT OF COMPLEMENT BY SERUM C'1 ESTERASE INHIBITOR

The fraction of human serum designated as C'1 esterase inhibitor is known to inhibit the action of C'1 esterase, a plasma kallikrein, and PF/Dil, an enzyme in plasma enhancing cutaneous vascular permeability. In the present study, C'1 esterase inhibitor has been found to block the actions of plasmin and the C'1r subcomponent of the first component of complement, and to retard the generation of ...

Studies on the Inhibition of Proteolytic Enzymes by Serum

Variation in the inhibition of trypsin, chymotrypsin, and plasmin by serum was studied in 65 normal individuals and in 117 patients with a variety of diseases. It was shown that elevated inhibition for trypsin and chymotrypsin is associated with disease processes that bring about cellular destruction. Changes in the inhibition of trypsin and chymotrypsin were closely correlated with changes in ...

Eotaxin promotes eosinophil transmigration via the activation of the plasminogen-plasmin system.

The effect of eotaxin, a potent eosinophil chemotactic factor, on eosinophil transmigration through a reconstituted basal membrane (Matrigel) was evaluated. Eotaxin induced significant eosinophil transmigration in the presence of 10% fetal bovine serum (FBS) and interleukin-5. Its effect was optimal at 0.01 microM, and it plateaued at 18 h. Eotaxin's effect was greater with eosinophils from ast...

Inhibition of trypsin, chymotrypsin, and plasmin by an inhibitor isolated from sheep serum.

A proteolytic enzyme inhibitor has recently been isolated from sheep serum and, by several criteria, viz. electrophoretic, chromatographic, and ultracentrifugal analysis, apparently behaved as a single component (1). Rat skin proteinase A (2) was used as the test enzyme for the purification of the inhibitor. It has now been determined that this inhibitor preparation can also decrease the activi...

The antiproteolytic activity of serum, and its changes in association with disease are little understood in spite of the extensive work that has been done in this field. The increase in inhibition by serum during various disease states has been repeatedly studied since the original observation of Ascoli and Bezzola in 1903 (1) (reviews, 2, 3). Trypsin has been the enzyme usually employed in the...
